Abstract
Salah satu sel yang biasa digunakan untuk berbagai penelitian kanker payudara in vitro adalah MCF-7. Beberapa penelitian mulai diarahkan pada pengujian potensi bahan alam yang berpotensi sebagai agen pendamping kemoterapi salah satunya adalah trisindolina. Oleh karena itu, pada penelitian tugas akhir ini dilakukan uji kinetika proliferasi sel untuk mengetahui aktifitas proliferasi sel kanker payudara MCF 7 cell line pada perlakuan trisindolina 1 menggunakan uji doubling time. Metode yang digunakan adalah dengan perlakuan pemberian doxorubicin dengan konsentrasi 1.536; 3.125; 6.25; 12.5; 25; 50; 100 µg/ml dan Trisindolina 1 dengan konsentrasi 1.536; 3.125; 6.25; 12.5; 25; 50; 100µg/ml. Lalu diuji sitotoksisitasnya dengan MTT-assay dan uji antiproliferatif dengan metode Doubling time. Hasil dari penelitian ini adalah senyawa trisindolina 1 memiliki aktivitas sitotoksisitas yang tinggi dengan IC50 15.773 µg/mL sehingga berpotensi sebagai senyawa anti kanker. Senyawa trisindolina 1 pada jam ke 24 sudahmampu menghambat proliferasi sel kanker payudara MCF-7 pada konsentrasi IC50 15.773 µg/mL.

One cell that is commonly used for various breast cancer studies in vitro is MCF-7. Several studies began to be directed at testing the potential of the material submitted as a chemotherapy companion agent, one of which is trisindolina. Therefore, in this final project a cell proliferation kinetics test was conducted to determine the cell line MCF 7 cell proliferation activity in the Trisindolina 1 consultation using a time doubling test. The method used is the composition of administration of doxorubicin with a concentration of 1,536; 3.125; 6.25; 12.5; 25; 50; 100 μg / ml and Trisindolina 1 with a concentration of 1,536; 3.125; 6.25; 12.5; 25; 50; 100μg / ml. Then submit the cytotoxicity with the MTT-assay and the antiproliferative test with the doubling time method. The results of this study are that trisindolina 1 has high cytotoxicity activity with IC50 15.773 μg / mL so that it can be developed as an anti-cancer ingredient. Trisindolina 1 compound at 24 hours has been able to inhibit proliferation of MCF-7 breast cancer cells at IC50 concentration of 15,773 μg / mL.
