Abstract
Atrazin merupakan herbisida yang bersifat persisten di lingkungan dengan waktu paruh sekitar 41-231 hari hingga bertahun-tahun. Pemakaian atrazin secara terus menerus pada tanah pertanian dapat menyebabkan penumpukan dan meninggalkan residu di tanah sehingga berpotensi mencemari tanah, air tanah dan permukaan air. Keberhasilan degradasi salah satunya dipengaruhi oleh faktor konsentrasi atrazin. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh interaksi konsentrasi atrazin dan lama waktu terhadap persentase degradasi dan viabilitas sel khamir. Pengukuran viabilitas sel dilakukan dengan menggunakan metode Total Plate Count (TPC) dan persentase degradasi menggunakan spektrofotometer. Kultur cair dengan konsentrasi atrazin 30 μg/L, 60 μg/L, 90 μg/L, 120 μg/L, 150 μg/L diinkubasi selama 8 hari dan dilakukan pengukuran tiap 2 hari. Penelitian didesain dengan menggunakan Rancangan Acak Lengkap (RAL) dengan tiga kali pengulangan. Data dianalisis secara statistik. Hasil penelitian menunjukkan bahwa konsentrasi atrazine dan lama waktu berpengaruh meningkatkan persentase degradasi dengan hasil degradasi tertinggi dicapai oleh konsentrasi 120 μg/L pada lama waktu hari ke 8 serta meningkatkan viabilitas sel dengan hasil jumlah total sel tertinggi dicapai oleh konsentrasi 30 μg/L pada lama waktu hari ke 4.

Atrazine is a recalcitrant herbicide in an environment with a half-life of about 41-231 days to years. Continuous use of atrazine in agricultural soils can cause accumulation and leave residues in the soil and potentially pollute the soil, ground water and surface water. One of the successes of degradation is influenced by atrazine concentration factors. The purpose of this research is to determine the effect of the interaction of atrazine concentration and incubation time on the percentage of yeast cell degradation and viability. Measurement of cell viability was carried out using the Total Plate Count (TPC) method and the percentage of degradation using a spectrophotometer. Liquid culture with atrazine concentration 30 μg/L, 60 μg/L, 90 μg/L, 120 μg/L, 150 μg/L was incubated for 8 days and measurements were taken every 2 days. The study was designed using a Completely Randomized Design (CRD) with three repetitions. Data were analyzed statistically. The results showed that atrazine concentration and incubation time increased the percentage of degradation with the highest result obtained by concentration of 120 μg/L at the time of day 8 and increased cell viability with the highest total cell counts achieved by a concentration of 30 μg/L at incubation of day 4.
