Abstract

Fosfat merupakan salah satu unsur makro esensial. Secara alami fosfat di dalam tanah berbentuk senyawa organik dan anorganik yang tidak larut, sehingga ketersediaannya di tanah sangat terbatas. Aktivitas mikroba tanah berpengaruh terhadap ketersediaan fosfat di dalam tanah melalui sekresi asam organik dan enzim fosfatase yang dihasilkan oleh mikroba sehingga fosfat tidak larut menjadi fosfat terlarut. Azotobacter telah diisolasi dari lahan Eco Urban Farming ITS. Isolat Azotobacter tersebut mampu melakukan metabolisme karbohidrat dan menghasilkan asam organik.
Skrining isolat yang berpotensi melarutkan fosfat secara kualitaif menggunakan medium Pikovskaya padat ditunjukkan dengan terbentuknya zona bening disekitar koloni. Tiga isolat yang memiliki Indeks Kelarutan Fosfat (IKF) terbesar diuji kemampuannya dalam melarutkan fosfat menggunakan medium pikovskaya cair.
Hasil skrining menunjukkan 8 isolat mampu membentuk zona bening. 3 isolat memilki nilai IKF yang relatif tinggi yaitu isolat A2, A5, dan A8. Isolat A2 mampu menghasilkan fosfat terlarut sebesar 31,9 ppm A5 sebesar 27,59 ppm, dan A8 sebesar 37,93 ppm. Ke-3 isolat menunjukkan hasil bahwa semakin lama waktu inkubasi konsentrasi fosfat terlarut yang dihasilkan semakin tinggi.

Phosphate is an essential macro elements. Phosphate in the soil form of organic and inorganic compounds are not soluble, so that availability in the soil is very limited. Soil microbial activity affects the availability of phosphate in the soil through organic acid secretion and phosphatase enzyme produced by microbes so that insoluble phosphate becomes soluble phosphate. Azotobacter has been isolated from the soil in Eco Urban Farming ITS. Azotobacter isolates are able to metabolize carbohydrates and produce organic acids.
Qualitative screening isolates using Pikovskaya solid medium, the formation of a clear zone around the colonies showed positive results in which isolates were able to dissolve the phosphate. Three isolates which have the largest phosphate solubility index (IKF) were tested for their ability to dissolve phosphate using Pikovskaya liquid medium.
Screening results proved that 8 isolates were able to set up a clear zone. Three isolates have the value of high IKF; isolates A2, A5, and A8. A2 isolates were able to produce soluble phosphate 31.9 ppm, A5 27.59 ppm and A8 37.93 ppm. All three isolates indicated that the longer the incubation time, the higher the concentration of dissolved phosphate generated.
