Abstract

Kadmium (Cd) yang diketahui sebagai salah satu logam berat berbahaya memiliki potensi untuk mencemari lingkungan dan menghambat pertumbuhan tanaman. Jewawut (Setaria italica) merupakan tanaman serealia alternatif pangan yang memiliki tingkat toleransi tinggi terhadap cekaman abiotik seperti kekeringan dan salinitas. Penelitian ini bertujuan untuk menguji pengaruh cekaman logam berat Cd terhadap respon morfo-fisiologi serta molekular tanaman Jewawut aks. Buru Merah yang dikultur secara hidroponik. Pada penelitian ini digunakan 4 taraf konsentrasi Cd (0, 0,5, 1,0 dan 1,5 µM pada media pertumbuhan ABmix) yang diberikan selama 4 minggu. Efek dari cekaman Cd yakni terganggunya pertumbuhan Jewawut aks. Buru Merah pada beberapa parameter morfo-fisiologi, terutama pada konsentrasi Cd tertinggi (1,5 µM) di usia panen 42 HST. Efek negatif dari cekaman Cd, seperti; penurunan tinggi tajuk, panjang akar, jumlah daun dan tunas, kandungan biomassa malai serta kadar klorofil total, meningkat seiring dengan peningkatan konsentrasi Cd yang diberikan pada media pertumbuhan Jewawut aks. Buru Merah selama 4 minggu. Berdasarkan kajian literatur, cekaman logam berat Cd menghasilkan MDA (malondialdehyde) yang lebih tinggi dan meningkatkan ekspresi genetik beberapa gen pengkode transporter logam berat seperti SiNRAMP, SiMTP, SiCAX, SiHMA, SiIRT dan SiZIP.

Cadmium (Cd) known as one of the dangerous heavy metals, has the potential to pollute the environment and inhibit plant growth. Foxtail millet (Setaria italica) is an alternative cereals food which have a high tolerance level to abiotic stresses such as drought and salinity. This study aims to examine the effects of Cd stress on the morpho-physiological and molecular responses of Foxtail millet accession Buru Merah that is cultured hydroponically. In this study used 4 levels of Cd concentrations (0, 0,5, 1,0 and 1,5 µM in ABmix growth media) given for 4 weeks. The effect of Cd stress was the growth disruption of Foxtail millet accession Buru Merah on several morpho-physiological parameters, especially at the highest Cd concentration (1,5 µM). The negative effects of Cd stress, such as; decreased of shoot length, root length, number of leaves and shoots, panicles biomass and chlorophyll content, increased along with the level of Cd concentration given to the growth media of Foxtail Millet accession Buru Merah. Based on literature, plants under Cd stress produce higher MDA (malondialdehyde) and have increased genetic expression in several heavy metal transporter coding genes such as SiNRAMP, SiMTP, SiCAX, SiHMA, SiIRT and SiZIP.
