Abstract
Karakter plastik yang sulit terdegradasi di lingkungan
mendorong penelitian ini untuk mengetahui potensi bakteri
Bacillus dan Pseudomonas koleksi Laboratorium Mikrobiologi
Biologi ITS dalam mendegradasi beberapa jenis plastik uji.
Metode yang digunakan adalah Kolom Winogradsky
dengan botol ukuran 1,5 l, berisi pasir steril : medium Salt = 1:1.
Plastik uji yang digunakan adalah kantong plastik merah, biru,
hitam, putih dan hijau ukuran 15 x 4 cm. Botol diisi 15 potongan
plastik. Proses biodegradasi dilakukan selama 4 bulan, dengan
parameter berat kering plastik, dan pengukuran densitas bakteri
uji pada λ 600 nm.
Isolat bakteri Bacillus dan Pseudomonas memiliki potensi
dalam mendegradasi plastik uji, karena mampu tumbuh dalam
kolom Winogradsky selama 4 bulan masa inkubasi. Isolat
Bacillus memiliki Optical Density pada plastik hitam : putih :
biru : hijau : merah sebesar 0,210: 0,234: 0,180: 0,183: 0,172.
Isolat Pseudomonas pada plastik hitam : putih : biru : hijau :
merah sebesar 0,196: 0,200: 0,160: 0,161: 0,176. Pada isolat
Bacillus persen degradabilitas plastik warna hijau, putih, hitam,
biru, merah secara berturut adalah 1,9%, 2,3%, 1,6%, 1,6%,
1,6%. Pada isolat Pseudomonas secara berturut adalah 1,7%,
2,2%, 1,4%, 1,6%, 1,7%

The characters of plastic that hard to be degraded in the
environment to encourage this study to determine the potential of
the bacteria Bacillus and Pseudomonas (ITS Biology
Microbiology Laboratory collection) in some types of plastics
test.
The method used is the Winogradsky column with a 1.5 l
bottle size, containing sterile sand: medium Salt = 1:1. Plastic test
used was a plastic bag of red, blue, black, white and green size 15
x 4 cm. Bottles filled with 15 pieces of plastic. Biodegradation
process carried out for 4 months, with a plastic dry weight
parameters, and measurements of bacterial density test at λ 600
nm.
Isolates of Bacillus and Pseudomonas bacteria have the
potential to degrade the plastic test, because it is able to grow in
the Winogradsky column for 4-month incubation period. Optical
Density of Bacillus isolates on the black plastic: white: blue:
green: red by 0.21: 0.234: 0.180: 0.183: 0.172. Pseudomonas
isolates on black plastic: white: blue: green: red by 0.196: 0.200:
0.160: 0.161: 0.176. In Bacillus isolates percent degradability of
green plastic, white, black, blue, red, respectively were 1.9%,
2.3%, 1.6%, 1.6%, 1.6%. In Pseudomonas isolates, respectively
were 1.7%, 2.2%, 1.4%, 1.6%, 1.7%
