Abstract
High Density polyethylene (HDPE) adalah termoplastik polietilen yang terbuat dari minyak bumi dan diproduksi secara komersial. HDPE memiliki sedikit percabangan, sehingga menghasilkan struktur yang lebih padat dan rapat daripada Low Density Polyethylene (LDPE). Hal ini yang menyebabkan mikroorganisme sulit untuk mengakses rantai polimer. Biodegradasi merupakan solusi paling efektif untuk mengatasi sampah plastik. Mikroorganisme yang bertindak sebagai agen biodegradasi yaitu konsorsium yeast dan mold Aspergillus terreus yang bekerja secara sinergis melalui pelekatan hifa dan pelepasan enzim. Aktivitas enzimatik yang dilakukan oleh mikroorganisme dalam mendegradasi menyebabkan menurunnya berat molekul dan hidrofobisitas polimer.
Uji biodegradasi dilakukan selama 30 hari dengan potongan plastik. Parameter hasil biodegradasi diantaranya yaitu efektivitas degradasi (%), jumlah total mikroba hetrotrofik (CFU/ml), dan analisis Scanning Electron Microscope (SEM). Perhitungan degradasi plastik dilakukan setiap 1 minggu sekali selama 4 minggu dengan menggunakan Rancangan Acak Lengkap (RAL). Analisis data menggunakan ANOVA-Two way dengan uji lanjut Duncan.
Hasil penelitian menunjukkan jenis konsorsium yeast dan mold dengan waktu inkubasi yang berbeda berpengaruh terhadap degradasi plastik HDPE yang ditunjukkan dari nilai efektivitas degradasi (%). Nilai tertinggi dicapai oleh kelompok perlakuan yang terdiri atas konsorsium yeast dan mold, tetapi tidak berbeda signifikan dengan yang terdiri dari konsorsium yeast. Jenis konsorsium yeast dan mold dengan waktu inkubasi yang berbeda berpengaruh terhadap jumlah total mikroba heterotrofik. Hasil uji SEM terlihat adanya formasi biofilm pada seluruh perlakuan yang menunjukkan adanya aktivitas biodegradasi plastik

High Density polyethylene (HDPE) is a polyethylene thermoplastic made from petroleum and produced commercially. HDPE has less branching, resulting in a denser and denser structure than Low Density Polyethylene (LDPE). This makes it difficult for microorganisms to access the polymer chains. Biodegradation is the most effective solution to overcome plastic waste. Microorganisms that act as biodegradation agents are yeast consortium and Aspergillus terreus that work synergistically through the attachment of hyphae and enzymes. The enzymatic activity carried out by microorganisms in degradation causes a decrease in the molecular weight and hydrophobicity of the polymer.
The biodegradation test was carried out for 30 days with plastic pieces. Parameters resulting from biodegradation include effectiveness of degradation (%), number of heterotrophic microbes (CFU/ml), and Scanning Electron Microscope (SEM). The calculation of plastic degradation was carried out once every 1 week for 4 weeks using a Completely Randomized Design (CRD). Data analysis using ANOVA-Two way with Duncan's further test.
The results showed that the type of yeast and mold consortium with different incubation times had an effect on the degradation of HDPE plastic as indicated by the value of the effectiveness of the degradation (%). The highest score was achieved by the treatment group consisting of yeast consortium and mold, but it was not significantly different from the treatment consisting of yeast consortium. The type of yeast consortium and mold with different incubations affected the number of heterotrophic microbes. The results of the SEM test showed that there was biofilm formation in all treatments which indicated the presence of plastic biodegradation activity.
