Abstract

Gynura pseudochina merupakan tanaman keluarga Asteraceae yang biasa digunakan sebagai tanaman obat di Asia Tenggara. G. pseudochina mengandung senyawa bioaktif yaitu flavonoid. Permintaan pasar yang tinggi menyebabkan perbanyakan tanaman dan upaya peningkatan metabolit sekunder tanaman tersebut penting dilakukan. Penelitian ini dilakukan untuk mengetahui kombinasi hormon IAA dan BAP untuk propagasi tanaman, serta mengetahui pengaruh elisitor MeJA terhadap kandungan flavonoid total dan antioksidan hasil aklimatisasi dengan media hidroponik. Penelitian ini dilakukan menggunakan nodus aksenik G. pseudochina dan didesain dengan rancangan acak lengkap (RAL). Kandungan flavonoid total dan antioksidan dianalisis menggunakan ekstrak metanol. Hasil penelitian menunjukkan bahwa kombinasi IAA 0,25 ppm BAP 3 ppm dapat menginduksi tunas sebesar 12±1,41a, respon pembentukan akar diinduksi oleh media MS 0, IAA 0,25 dan IAA 0,5 ppm. Hasil elisitasi MeJA 100 µM menghasilkan kadar flavonoid terbesar yaitu 131,42±0,26c mgQE/DW dengan aktivitas antioksidan yang juga meningkat dibandingkan kontrol dengan nilai IC50 173,28 ppm. Selain itu, ditemukan pigmentasi pada umbi setelah treatment MeJA 50 µM dan 100 µM G. Pseudochina yang mengindikasikan adanya akumulasi senyawa metabolit sekunder.

Gynura pseudochina is a plant of the Asteraceae family which is commonly used as a medicinal plant in Southeast Asia. G. pseudochina contains bioactive compounds, namely flavonoids. The high market gap causes plant propagation and efforts to increase secondary metabolites of these plants are important. This study was conducted to determine the combination of IAA and BAP hormones for plant propagation, and to determine the effect of the elicitor MeJA on TFC and antioxidant activity of acclimatized using hydroponic media. This study was conducted using the accentic node of G. pseudochina and was designed with a completely randomized design (CRD). Total flavonoid content and antioxidants were analyzed using methanol extract. The results showed that IAA 0,25 ppm and BAP 3 ppm could induce shoots of G. Pseudochina about 12±1,41a and the root able to induce using MS 0 , IAA 0,25, and IAA 0,5 ppm medium. The elicitation of MeJA 100 M produced the largest flavonoid content, namely 131.42±0.26c mgQE/DW meanwhile antioxidant activity increased compared to control with IC50 value of 173.28 ppm. In addition, the pigmentation of the tubers after treatment with MeJA 50 M and 100 M G. Pseudochina was found to be an accumulation of secondary metabolites.
