Abstract
Pencemaran akibat limbah pewarna sintetik merupakan salah satu permasalahan yang dihadapi di berbagai negara. Salah satu jenis pewarna sintetik yang berbahaya yaitu Reactive Black 5 (RB5) karena bersifat beracun dan karsinogenik. Jamur Gloeophyllum trabeum (GT) memiliki potensi untuk mendegradasi pewarna sintetik dengan adanya enzim-enzim dan reaksi Fenton yang dimiliki. Pada penelitian ini, jamur GT dikombinasi dengan Metal Organic Framework (MOF) UiO-66 dan material pengimobilisasi polivinil alkohol (PVA) dan natrium alginat (sodium alginat, SA) untuk mendegradasi pewarna sintetik RB5. Dua jenis komposit telah berhasil diproduksi dari kombinasi ini, biokomposit pertama UiO-66@GT didapat dengan menambahkan variasi UiO-66 (15, 30, 45 mg) pada media pertumbuhan kultur GT. Biokomposit kedua UiO-66/GT@PVA-SA diperoleh dengan mengimobilisasi kultur GT dengan UiO-66 yang telah dihomogenasi bersama campuran PVA-SA (4% dan 2% b/v). Campuran tersebut diteteskan perlahan ke dalam larutan dingin asam borat dan CaCl2 yang distirer untuk mendapatkan beads. Hasil uji dekolorisasi RB5 dengan UiO-66@GT diperoleh persentase dekolorisasi 72,55%, sedangkan kultur GT murni hanya mencapai 36,47%. Sementara untuk UiO-66/GT@PVA-SA, dekolorisasi RB5 mampu mencapai 80,12%. Uji kinetika adsorpsi RB5 dengan komposit UiO-66@GT dan UiO-66/GT@PVA-SA menunjukkan bahwa proses adsorpsi mengikuti model kinetika orde dua semu dengan koefisien korelasi R2 masing-masing 0,9849 dan 0,9997. Kedua komposit juga menunjukkan proses adsorpsinya bersifat spontan dan feasible dari uji termodinamika adsorpsinya dengan nilai ∆G yang negatif. Hasil identifikasi produk metabolit dekolorisasi RB5 dengan biokomposit UiO-66/GT@PVA-SA menggunakan alat LC-QTOF MS diusulkan 4 metabolit yaitu: C24H19N5Na2O13S4 (m/z = 762), C10H13N2O8S2- (m/z = 353), C12H9N4O7S2- (m/z = 384), dan C10H13O8S2- (m/z = 325). Hasil-hasil tersebut membuktikan bahwa biokomposit UiO-66@GT dan UiO-66/GT@PVA-SA memiliki potensi untuk mendekolorisasi pewarna sintetik seperti RB5.

Synthetic dye waste pollution is one of the problems faced in various countries. One type of dangerous synthetic dye is Reactive Black 5 (RB5) because it is toxic and carcinogenic. The fungus Gloeophyllum trabeum (GT) is the potential fungus that has ability to degrade synthetic dyes in the presence of its Fenton reactions and enzymes. In this study, GT fungus were combined with UiO-66 Metal Organic Framework (MOF) and immobilized in polyvinyl alcohol (PVA) and alginate (sodium alginate, SA) matrix which used to degrade synthetic RB5 dye. Two kinds of composites have been successfully synthesized from this combination, the first biocomposite UiO-66@GT was obtained by adding variations of UiO-66 (15, 30, 45 mg) to GT culture growth medium. The second one, UiO-66/GT@PVA-SA was gained by immobilizing GT culture with UiO-66 which had been homogenized with the PVA-SA mixture (4% and 2% w/v). The mixture was dripped slowly into a cold solution of boric acid and CaCl2 which was stirred to get beads. The results of the RB5 decolorization test with UiO-66@GT obtained a decolorization percentage of 72.55%, while pure GT culture only reached 36.47%. While UiO-66/GT@PVA-SA, RB5 decolorization was able to reach 80.12%. The RB5 adsorption kinetics test with the UiO-66@GT and UiO-66/GT@PVA-SA composites showed that the adsorption process followed a pseudo second order kinetics model with correlation coefficients (R2) 0.9849 and 0.9997, respectively. The two composites also showed that the adsorption process was spontaneous and feasible from the results of the adsorption thermodynamic test with negative ∆G value. The results of the identification of decolorization metabolite products with the UiO-66/GT@PVA-SA biocomposite using the LC-QTOF MS suggested 4 metabolites, namely: C24H19N5Na2O13S4 (m/z = 762), C10H13N2O8S2- (m/z = 353), C12H9N4O7S2- (m/z = 384), dan C10H13O8S2- (m/z = 325). These results proved that UiO-66@GT and UiO-66/GT@PVA-SA biocomposites have the potential to decolorize synthetic dyes such as RB5.
